背景

Buffer Exchange 是進行樣品製備與穩定性測試的必經過程，不管是Desalting 或是更換反應環境，也是最枯燥費時的步驟．緩衝液交換失誤，可能造成短暫的環境改變，如pH改變, 過度濃縮甚至換錯buffer，都可能造成樣品聚集甚至結構上的改變，而影響後續結果．Big Tuna& Unagi可以幫你行全自動化的Buffer exchange流程，並且提估更精細的控制與監控。

液體置換、樣本濃縮首選Big Tuna ＆Unagi

傳統透析法（Dialysis）是最簡單、廣泛使用的方式之一，但是殘留大且要求實驗技術，速度也是個重要的考慮因素；Centrifugation，或是真空吸引是快速的方式，但是此方法一定會造成短暫的濃度大幅升高，且有蛋白質變性或卡管的問題：切向流過濾(Tangential Flow Filtration, TFF)相較來說是一種快速且不會造成短暫濃度上升造成的極化現象的方式，但其前置備時間長且需要樣品量大才符合經濟效應。

而Big Tuna & Unagi皆採用UF/DF的方式進行buffer exchange, 並且在每次加完buffer與過濾後都會利用超音波偵測器進行液面高度量測．不僅可以監控交換過程；也可以控制最高的濃縮率，例如可以控制每次僅交換50%的新緩衝液，重複交換10次便可以達到1%以下的原緩衝液．採用正壓式過濾，並搭配mixing搖動的功能，壓力與搖動速度皆可控制．採用最受控的方式對待你最珍貴的樣品．
