MACS Tissue Storage Solution

在研究上常常需要處理新鮮組織，但有時無法立刻進行下一步操作。為了讓組織解離過程更靈活，同時保證細胞產出和活力，Miltenyi Biotec 開發了組織儲存液。這項革命性的解決方案能將新鮮器官和組織樣本保存長達 48 小時，期間細胞活力完全不受影響，也不會出現像細胞活化或細胞凋亡等不想要的副作用。組織儲存液已經過多種人體和小鼠組織（包括腫瘤、皮膚、心臟、脾臟、大腦和骨骼肌）的測試和驗證，安全性高效值得信賴。(研究中經常需要儲存新鮮器官和實體組織樣本，例如樣品需要運送至外部機構再運回，或者在不同時間採集，又或者無法立即全部處理。這時候，短期儲存能夠在幾天內為組織的進一步處理提供充足的靈活性。分析儲存組織中的活細胞時，避免細胞在儲存期間發生壞死和凋亡至關重要。此外，還必須防止細胞活化和多能性的改變，以保持細胞的生理和功能狀態。冷凍組織樣本是一種常見的儲存方法，但解凍後通常會導致細胞死亡率升高，並且運輸也存在挑戰。為了克服這些問題，我們專門開發了 MACS 組織儲存液，可以在 2-8°C 的條件下將新鮮實體組織至少保存 48 小時，並使其處於最佳狀態。在此儲存期間，細胞活力、功能和啟動狀態得以保持，同時可以防止細胞應激相關基因的誘導。MACS 組織儲存液已在多種人類和小鼠組織上進行了測試，包括腫瘤、肺、脾臟、大腦和骨骼肌。

Tissue dissociation kits

MACS® 組織解離試劑盒，能從幾乎任何類型的實體組織中，高效獲取高產量的保存了表位完整性的活單細胞。我們優化的方案結合了機械解離和酶處理，能取得具有出色活力的單細胞懸浮液，同時提供高度標準化及具重複性的結果，減少人為誤差。

MACS® SmartStrainers

MACSmix Rotator

MACSmix™ 管式旋轉器是一款多功能儀器，可使用可充電電池獨立運行，無需固定電源。此外，也可以連接電源使用。它適用於 2°C 至 42°C 的溫度範圍，因此可以放置在冰箱或培養箱中。此外，其可有效混合各種試管尺寸的樣品，適用於 0.5 ml至 50 ml的試管。該儀器可以三種不同的速度或按設定間隔運行。

Removal reagents

此試劑是用於清除細胞懸浮液中死亡細胞和有害物質的試劑。細胞懸浮液中的有害物質會干擾各種下游應用，例如細胞分離、細胞分析或細胞定序。我們的清除試劑可以通過有效去除死亡細胞、碎片、紅細胞、髓鞘或內毒素，來優化您樣本的製備，從而獲得可靠的結果。以下為可選擇使用的套件:

• Dead Cell Removal kit: 死細胞去除試劑盒是一種從細胞培養物或組織製劑中去除死細胞的快速、直接的方法。死細胞去除試劑盒包含即用型微珠和結合緩衝液，用於對細胞碎片、死細胞和垂死細胞進行磁性標記。透過磁力分離去除磁性標記材料，並在 25 分鐘內獲得純淨的活細胞。
• Debris Removal Solution: 是一款即用型密度梯度試劑。它可在離心過程中使用全速啟動和停止，快速有效地從各種組織類型的解離細胞中去除細胞碎片，從而保留活細胞。
• Red Blood Cell Lysis Solution (10x): 專為優化紅血球裂解而開發，可在人、小鼠或大鼠全血、骨髓以及組織製備的單細胞懸浮液中高效裂解紅血球，且對其他細胞的影響最小。如果流式細胞儀分析在裂解後超過一小時進行，建議在細胞分離或分析前進行一次洗滌步驟。
• Myelin Removal Beads II, human, mouse, rat: 可在樣品製備期間從單細胞懸浮液中特異性去除髓鞘碎片，從而提高目標細胞的純度和回收率，從而極大地改善了後續的細胞分離和抗體染色。因此，建議在細胞分選或抗體染色之前進行髓鞘去除。
• Endotoxin Removal Beads: 是從純化的蛋白質製劑中快速、輕鬆去除內毒素和 DNA 的理想工具。
• Basic MicroBeads: 是未經偶聯的微珠。它們可以用於去除非特異性地結合到微珠上的物質，例如從樣品中去除粘性細胞或死細胞，它們也用於在角質形成細胞內吞作用後分離溶酶體。

FcR Blocking Reagents

MACS Tissue Storage Solution

Oftentimes tissue samples cannot be processed immediately when they become available. To enable flexible timing of tissue dissociation without compromising cell yield or viability, Miltenyi Biotec developed the Tissue Storage Solution. This solution enables storage of fresh organ and tissue samples for up to 48 hours. During this time, cell viability is not affected and no undesired effects like cell activation or apoptosis induction have been observed. Tissue Storage Solution was tested and validated on a variety of human and mouse tissues including tumors, skin, heart, spleen, brain, and skeletal muscle.

Tissue dissociation kits

MACS Tissue Dissociation Kits and gentleMACS Dissociators and Tubes are perfectly geared to each other. The combination of gentle mechanical and enzymatic tissue dissociation preserves all cell characteristics and functions for reliable and meaningful downstream analysis. Pre-defined enzyme solutions and lot-to-lot consistency of enzyme activities ensure reproducible and user-independent results.

Miltenyi Biotec researchers tested whether the proteases included in the Tumor Dissociation Kits for human and mouse tissue have an effect on the integrity of cell surface proteins. To this end, the presence of more than 200 epitopes was analyzed by flow cytometry after incubation of cells with the enzymes. The vast majority of the cell surface epitopes tested was well preserved during enzymatic treatment.

MACS® SmartStrainers

Cell strainers and filters in various mesh sizes for rapidly removing cell aggregates and tissue fragments from blood samples or cell suspensions after tissue dissociation. The versatile MACS® SmartStrainers and Pre-Separation Filters are the best solution for the preparation of uniform single-cell suspensions for downstream uses, like in cell separation columns or in flow cytometers.

MACSmix™ Tube Rotator

The MACSmix™ Tube Rotator is a versatile instrument running on rechargeable batteries and operating independently of a permanent power supply. Alternatively, mains operation is possible. It is suitable for a temperature range of 2 °C to 42 °C and can, therefore, be placed in a refrigerator or incubator.The MACSmix Tube Rotator provides effective overhead sample mixing in various tube sizes from 0.5 mL to 50 mL tubes. The device operates at three different speeds or at set intervals.

Removal reagents

Unwanted material in cell suspensions can have interfering effects in different downstream applications, such as cell separation, cell analysis, or cell sequencing. Our removal reagents help you optimize your sample preparation by enabling the efficient removal of dead cells, debris, red blood cells, myelin, or endotoxins to get reliable results.

FcR Blocking Reagent

 Incubation of cells with FcR Blocking Reagent, mouse, increases the specificity of labeling with MACS® Antibodies or MACS MicroBeads and thereby improves the specificity of immunofluorescent staining and purity of magnetically isolated target cells, respectively.
